COMBINED ALCIAN BLUE - P.A.S

SECTIONS
4micron paraffin wax sections

SOLUTIONS

1. Alcian Blue pH 2.5

  • Alcian Blue 1g
  • 3% Acetic Acid 100ml

2. 1% Periodic Acid

3. Schiffs Reagent

METHOD

  1. Take sections to water
  2. Place in Alcian Blue for 5 minutes
  3. Wash in distilled water
  4. Place in 1% periodic acid for 10 minutes
  5. Rinse well in distilled water
  6. Place in Schiffs reagent for 20 minutes
  7. Wash in running tap water for 10 minutes
  8. Stain lightly in Gills Haematoxylin
  9. Wash and 'blue' in 1% Lithium carbonate
  10. Dehydrate, clear . Mount sections in DPX

RESULTS

  • Acid Mucins - Blue
  • Neutral Mucins - Magenta
  • Nuclei - Pale Blue
