Tissue Stain for Amyloid

HIGHMANS CONGO RED

SECTIONS

4µm paraffin wax sections


SOLUTIONS

a) 0.5% Congo Red in 50% alcohol.

b) 0.2% Potassium Hydroxide in 80% alcohol.


METHOD

  1. Sections to water
  2. Stain in congo red solution for 10-15 minutes
  3. Differentiate with alcoholic potassium hydroxide solution if necessary
  4. Wash in water, stain nuclei in Gills Haematoxylin, differentiate and blue
  5. Dehydrate, clear . Mount sections in DPX

RESULTS

  • Amyloid, elastic fibres, eosinophil granules - Red
  • Nuclei - Blue
