CROSS-PALMGRENS METHOD

SECTIONS

8 micron paraffin wax sections


SOLUTIONS

1. Reducer

  • Pyrogallol 10g
  • Distilled water 450ml
  • Absolute alcohol 550ml
  • 1% Nitric acid 2ml

2. 1% ammonia

3. Silver Solution

  • Silver nitrate 15g
  • Potassium nitrate 10g
  • Distilled water 100ml
  • 5% Acetic acid 1ml

4. 0.2% Gold Chloride

5. 5% Sodium Thiosulphate


METHOD

  1. Place reducer in 60C incubator for use at stage 5
  2. Take sections to water
  3. Place in 1% ammonia for 5 minutes
  4. Rinse in distilled water
  5. Impregnate with silver solution for 30-45 minutes
  6. Drain excess silver solution from the slides and without rinsing, pour on the warm reducer until the sections turn brown. Slight agitation prevents deposit formation
  7. Wash in distilled water
  8. Tone in 0.2% gold chloride for 2 minutes
  9. Wash in water
  10. Fix in 5% sodium thiosulphate solution
  11. Wash in water
  12. Dehydrate, clear . Mount sections in DPX

  • Neurofibrillary Tangles - Black
  • Senile plaque neurites - Black
  • Background - Grey
