GRIMELIUS

1. Sections to water.

2. Treat sections at 65ªc with silver solution for 3 hours

3. Drain silver solution from slide.

4. Treat with freshly prepared reducing solution at 45ªc for 1 minute.

5. Wash in distilled water

6. Repeat step 4 until the desired result is achieved.

7. Wash in distilled water.

8. Dehydrate, clear . Mount sections in DPX

--Argyrophil material black

--Background yellow


SOLUTIONS

SILVER STAINING SOLUTION

-Acetate buffer ph 5.6 10 mls

--Distilled water 87 mls

--1% silver nitrate 3 mls

REDUCING SOLUTION

--Sodium sulphite (hydrated) 2.5 g

--Hydroquinone (quinol) 0.5 g

--Distilled water 50 mls

ACETATE BUFFER pH 5.6

--0.2M acetic acid 4.8 mls

--0.2M sodium acetate 45.2 mls

--Distilled water 50 mls
