Stain for Basement Membrane and Mesangium

JONES METHENAMINE SILVER

1. Sections to distilled water.

2. Treat with 1% periodic acid for 10 minutes.

3 Wash in distilled water.

4. Place in hexamine silver solution pre-heated to 56ªc for 30-90 minutes controlling microscopically

5. Wash in distilled water.

6. Tone in 0.2% gold chloride

7. Wash in distilled water.

8. Counterstain in 1% light green or with a light haematoxylin and eosin.

9. Wash in water.

10. Dehydrate, clear . Mount sections in DPX


RESULTS

--Basement membrane and Mesangium black

--Background red or green --Background colour is dependant on the counterstain used


SOLUTIONS

HEXAMINE SILVER

--3% hexamine 20 mls

--5% silver nitrate 1 ml

--Mix until the precipitate disappears. Then add to:

--5% borax (sodium tetraborate) 2 mls

--Distilled water 25 mls

THIS SOLUTION MUST BE FILTERED BEFORE USE.


SAFETY

Waste ammoniacal silver solution is potentially explosive. Collect all ammoniacal silver and put into the silver waste bottle - it contains saturated sodium chloride to precipitate out the silver, this not only makes it safe but allows the silver to be sold for reclaimation.Do not pour waste solutions down the drain.
