Stain for fibrin

M.S.B TECHNIQUE

1. Sections to water.

2. Treat with iron alum for 5 minutes.

3. Wash water.

4. Stain with mayers haematoxylin for 1 minute, differentiate if necessary.

5. Blue in tap water.

6. Rinse in alcohol.

7. Stain with 0.2% orange g in alcoholic picric acid for 5 minutes

8. blot dry.

9. Stain with brilliant crystal scarlet solution for 3 minutes.

10. Wash in water.

11. Treat with phosphotungstic acid until the connective tissue appears pink/colourless approx 20-30 seconds.

12. Wash in water.

13. Stain with 1% aniline blue in 2% acetic acid for 2 seconds

14. Dehydrate, clear . Mount sections in DPX


RESULTS

--Nuclei blue/black

--Muscle red

--Erythrocytes red

--Collagen blue

--Fibrin red

--Early fibrin may be yellow

--Very old fibrin may be blue
