HAEMATOXYLIN AND EOSIN
1. Stain in Gill's double strength haematoxylin 10-20 minutes
2. Wash in distilled water.
3. Blue in Scotts tap water substitute.
4. Wash in water for 15 minutes.
5. Stain in 1% eosin in filtered 1% calcium chloride for 2-4 minutes.
6. Wash in water for 30 seconds.
7. Blot dry.
8. Wash in 100% alcohol for 20 seconds.
9. Rinse in xylene.
10. Mount in DPX.
RESULTS
--Nuclei Blue
--Cytoplasm Pink
--Muscle Red
--Collagen Pink
--RBCs Orange/red
GILLS DOUBLE STRENGTH HAEMATOXYLIN
--Haematoxylin 4g
--Distilled water 730ml
--Ethylene glycol 250ml
--Aluminium sulphate (Hydrated) 35g
--Acetic acid 20ml
Then add sodium iodate 0.4g
SCOTT'S TAP WATER SUBSTITUTE
--Potassium bicarbonate 1g
--Magnesium sulphate 10g
--Distilled water 500ml
PERIODIC ACID SCHIFF (PAS)
1. Treat solutions with 1% periodic acid for 20 minutes.
2. Rinse in distilled water.
3. Treat sections with Schiff's reagent for 30 minutes.
4. Wash in distilled water for 20 minutes.
5. Counterstain in Gill's haematoxylin for 20 seconds.
6. Blue.
7. Wash in water.
8. DCM.
RESULTS
-PAS positive material Magenta
--Nuclei Blue
