Stain for Hepatitis B and Copper Associated protein

SHIKATA TECHNIQUE

1. Sections to water.

2. Treat with acidified potassium permanganate for 3 minutes.

3. Wash in water.

4. Decolourize with 1% oxalic acid.

5. Wash well in water.

6. Stain with shikata orcein for 10-90 minutes.

7. Wash in water.

8. Dehydrate, clear . Mount sections in DPX


RESULTS

--Hepatitis B surface antigen,

--Copper associated protein,

--Elastin fibres purple/brown.


SOLUTIONS

SHIKATA ORCEIN --Orcein 1 g

--70% ethanol 100 mls

--Conc. hydrochloric acid 1 ml
